System description
The Aquilos is a cryo dedicated DualbeamTM system specifically designed to delivers optimal cryo sample preparation within the Thermo Scientific's Cryo-tomography workflow.
The Cryo-tomography workflow consists of a plunge freezing device (VitrobotTM), import- and correlative functionality of (cryo) light microscopic images via Maps software and high-end cryo-TEM systems i.e. Titan Krios and Talos Arctica. The Aquilos sits between the light microscope used for targeting regions of interest after freezing and the Titan Krios which is used for Cryo-TEM tomography at very high resolution.
The stage is cooled at <-180 °C and all samples are handled and loaded/unloaded at <-186 °C. LN2 is used as the cryogen and N2 is used as the carrier in the cooling loop: it is passed through a heat exchanger within a Dewar of LN2 and subsequently cools the stage before being vented outside of the specimen chamber (the LN2 boil-off rate is increased because of this cooling).
The loading stations are where samples are loaded and unloaded, and are essentially open Dewars of LN2.
User maintenance
Allow only trained personnel to perform user maintenance procedures. Read and understand the safety precautions throughout this manual. Always observe appropriate industry-approved safety methods and procedures.
Never attempt maintenance or service of any kind, other than that described for the user (see the appropriate User Manual).
There may be additional limitations to safety and handling not mentioned in this manual. If you have any doubt regarding approved safety procedures, contact safety personnel at your company, or representatives of your state, territory or province, or federal government.
General rules
• Do not maintain alone.
• Do not perform internal maintenance or adjustments unless you are a trained service person.
• Do not maintain unless another person, capable of rendering the first aid or resuscitation, is in the immediate vicinity.
• Always ensure a safe environment when performing any maintenance.
• Make sure you are familiar with the hazards of working with cryogenic asphyxiant gasses.
• Be aware of the location of the nearest phone and of the emergency service numbers.
• Make sure your hands are dry and that you are standing on a dry, insulated surface, capable of withstanding the accessed voltages.
• Make sure to clean your hands after maintenance.
